Synthesis of Novel Photoactive Enkephalin-Like Peptides

Abstract

Two novel photoaffinity analogues of enkephalin containing p-[3-(trifluoromethyl)-3H-diazirin-3-yl]-L-phenylalanine or p-azido-L-phenylalanine residues in position 4 ([D-Ala2, Phe(Tmd)4, D-Leu5]-enk, I, and [D-Ala2, Phe(N3)4, D-Leu5]-enk, II) were synthesized by classic technique in solution. Photochemical properties of the analogue I and II were studied under UV irradiation at 350 nm. Optimal time of photolysis for ligand-receptor complex was determined. Binding properties of the analogues with opiate receptors were investigated. In the [H3]DAGO and [H3]DSLET binding assays the analogue I showed almost the same affinity for m-receptor and the lower affinity for d-sites compared to the parent DADLE, while the analogue II retained affinity and preference for m-receptor.
