The Coenzyme Regeneration for Biosynthesis of Chiral Compounds Using Dehydrogenases

Abstract

Principles of synthesis of chiral compounds using dehydrogenases as the main biocatalyst and formate dehydrogenase as biocatalyst for coenzyme regeneration have been discussed. Process of preparation of new generation of biocatalysts for NAD(P)H regeneration using formate dehydrogenase (FDH, EC 1.2.1.2) from Pseudomonas sp.101 have been described. New recombinant E.coli strains producing different forms of FDH have been constructed and efficient and cheap procedure for enzyme purification have been developed. Protein engineering approaches have been used to prepare different forms of FDH with improved kinetic properties, increased thermal and chemical stability and changed coenzyme specificity.
