UV/Vis-spectrophotometric study of the interaction of proflavine with a-chymotrypsin in water-acetonitrile mixtures

Abstract

In present work the binding of proflavine with bovine pancreatic a-chymotrypsin (CT) in various water-acetonitrile mixtures has been measured by means of UV/Vis-spectrophotometry at 25oC. Thermodynamic activity of water in organic solvent was varied from 0.11 to 1.0. The spectrophotometric results on proflavine binding have been analyzed in comparison with literature results on corresponding changes in the protein conformational mobility and FTIR-data on water state in acetonitrile.
