Kinetics of quercetin oxidation catalyzed by horseradish peroxidase in the presence of b-cyclodextrin

Abstract

The rate of quercetin oxidation catalyzed by horseradish peroxidase was found to increase by 20% in the presence of macrocyclic complexing agent b-cyclodextrin. The comparison of the kinetic parameters of this reaction in the presence and in the absence of b-cyclodextrin showed that the introduction of b-cyclodextrin did not influence significantly the specificity of the enzyme towards the reducing substrate (estimated as kcat/Km ratio). At the same time, the reaction rate did not depend on the time of incubation of quercetin and b-cyclodextrin. The increase of the reaction rate was supposed to be associated with non-specific interaction between b-cyclodextrin and the product of quercetin oxidation.
