New system for TNF-alpha quantification in clinical samples

Abstract

Full-length TNF-alpha and its truncated derivative with 18 N-terminal amino acids replaced to T7-tag were produced in E. coli using artificial genes and rendered inclusion bodies. Purification and refolding methods were established for the recombinants, their biological activity was demonstrated. Polyclonal antibodies against TNF-alpha were raised. ELISA-Sandwich test-system for TNF-alpha quantification with 100 pg/ml sensitivity was engineered. The system was shown to be applicable for detection of the TNF-alpha elevation in human blood.
